Effectiveness of combining va-mediated resistance with a lettuce mosaic virus coat protein gene and a potato virus Y polymerase RNA gene in protection of tobacco hybrids from Potato virus Y

Growing resistant cultivars is the most effective method of plant protection against viral diseases. Different sources of resistance are used for breeding new tobacco cultivars resistant to Potato virus Y (PVY). The main source of resistance to the virus used in breeding is va-mediated resistance caused by a deletion within a susceptibility gene. Such a mutation is present in cultivar Virgin A Mutant (VAM) and similar resistance type is observed in case of Polish cultivar Wiślica. Other sources of resistance used in this study were transgenic tobacco breeding lines previously obtained by transformation with Agrobacterium: MN 944 LMV (containing lettuce mosaic virus coat protein gene LMV CP) and AC Gayed ROKY2 (containing potato virus Y polymerase RNA antisense gene ROKY2). Expecting that combination of two different sources of resistance in one genome will increase the resistance to PVY, we crossed cultivars VAM and Wiślica with lines MN 944 LMV and AC Gayed ROKY2. The following generations of offspring were tested for the presence of transgenes by PCR analyses. Transgenic hybrids were also tested for PVY resistance using two different isolates in artificial inoculation under greenhouse conditions. The resistance of tested plants was confirmed by serological tests with the use of specific antibodies. Crossing Wiślica and AC Gayed ROKY2 showed to be adverse. The most effective combination of va factor and transgene LMV CP was observed for VAM x MN 944 LMV and Wiślica x MN 944 LMV because a complete resistance was maintained in subsequent generations in the case of these hybrids. Finally, we received lines with combination of different sources of resistance which showed high level of resistance against PVY.
